The Important Role of Poly ADP-Ribose Polymerase Inhibitor in Prostate Cancer

نویسندگان

چکیده

Poly (ADP-ribose) polymerase inhibitors (PARP inhibitor) is a new targeted drug for metastatic prostate cancer in which the patient has mutation on homologous repair gene including BRCA1/2 gene. This was first gene-based agent and there were more than 5 available drugs PARP inhibitors. Only 4 approved clinical use patients with from U.S. Food Drug Administration. review article deals overview of their future research fields.

Poly (ADP-ribose) polymerase inhibitor: an evolving paradigm in the treatment of prostate cancer

Recent phase I studies have reported single-agent activities of poly (ADP-ribose) polymerase (PARP) inhibitor in sporadic and in BRCA-mutant prostate cancers. Two of the most common genetic alterations in prostate cancer, ETS gene rearrangement and loss of PTEN, have been linked to increased sensitivity to PARP inhibitor in preclinical models. Emerging evidence also suggests that PARP1 plays an...

The promising role of poly(ADP-ribose) polymerase inhibitors in prostate cancer

reported in the New England Journal of Medicine, was a targeted, biomarker, open-label, single-group, multi-site phase II trial design mostly in the United Kingdom, looking at the utility of olaparib in those who harbor deleterious germline BRCA2 mutations. The TOPARP trial enrolled a cohort of 45 mCRPC patients in this two-stage design (30 patients in the first cohort and 15 patients in the se...

Novel Poly(Adenosine Diphosphate-Ribose) Polymerase (PARP) Inhibitor, AZD2461, Down-Regulates VEGF and Induces Apoptosis in Prostate Cancer Cells

Background: Prostate cancer (Pca) is a heterogeneous disease, and current treatments are not based on molecular stratification. Poly(adenosine diphosphate [ADP]-ribose) polymerase (PARP) inhibitors have recently been found to be remarkably toxic to cells with defects in homologous recombination, particularly cells with BRCA-mutated backgrounds. Therefore, this preliminary study was designed to ...

Trapping Poly(ADP-Ribose) Polymerase.

Recent findings indicate that a major mechanism by which poly(ADP-ribose) polymerase (PARP) inhibitors kill cancer cells is by trapping PARP1 and PARP2 to the sites of DNA damage. The PARP enzyme-inhibitor complex "locks" onto damaged DNA and prevents DNA repair, replication, and transcription, leading to cell death. Several clinical-stage PARP inhibitors, including veliparib, rucaparib, olapar...

The role of poly(ADP-ribose) polymerase-1 inhibitor in carrageenan-induced lung inflammation in mice.

Increasing indication is unveiling a role for poly(ADP-ribose) polymerase (PARP)-1 in the regulation of inflammatory/immune responses. The aim of the present study was to determine the potential anti-inflammatory effects of PARP-1 inhibitor 5-aminoisoquinolinone (5-AIQ) to explore the role of PARP-1 inhibitor in a mouse model of carrageenan-induced lung inflammation. A single dose of 5-AIQ (1.5...
